The sodium leak channel NALCN regulates cell excitability of pituitary endocrine cells

چکیده

Anterior pituitary endocrine cells that release hormones such as growth hormone and prolactin are excitable fire action potentials. In these cells, several studies previously showed extracellular sodium (Na+) removal resulted in a negative shift of the resting membrane potential (RMP) subsequent inhibition spontaneous firing potentials, suggesting contribution Na+ background conductance. Here, we show leak channel NALCN conducts Ca2+- Gd3+-sensitive TTX-resistant conductance GH3 cell line, model cells. knockdown hyperpolarized RMP, altered electrical properties inhibited secretion. Conversely, overexpression depolarized also reshaping Overall, our results indicate is functional involved excitability well Indeed, line suitably models native display similar appears proper cellular to study role excitability.

The sodium leak channel, NALCN, in health and disease

Ion channels are crucial components of cellular excitability and are involved in many neurological diseases. This review focuses on the sodium leak, G protein-coupled receptors (GPCRs)-activated NALCN channel that is predominantly expressed in neurons where it regulates the resting membrane potential and neuronal excitability. NALCN is part of a complex that includes not only GPCRs, but also UN...

Roles of the Ion Channel Nalcn in Neuronal Excitability Control

The resting membrane potential (RMP) of a neuron is set by a complex balance between charged ions, ion channels and transporters. Many of the ion channels have been identified at the molecular level. Missing from the molecular identification has been the voltage-insensitive background sodium ‘‘leak’’ conductance that depolarizes the RMP from the equilibrium potential of potassium and provides a...

Sodium Leak Current through a Non-selective Cation Channel Regulates Spontaneous Activity of Pacemaker Cells

Pacemaker cells are involved in regulating numerous essential biological functions. These cells exhibit spontaneous activities in isolation, due to the various membrane ion channels. Background conductances (ILeak) are responsible for the depolarized resting membrane potential of numerous pacemaker cells, largely due to a high sodium component (INa Leak); however, their molecular identity remai...

The NALCN ion channel is activated by M3 muscarinic receptors in a pancreatic β-cell line

A previously uncharacterized putative ion channel, NALCN (sodium leak channel, non-selective), has been recently shown to be responsible for the tetrodotoxin (TTX)-resistant sodium leak current implicated in the regulation of neuronal excitability. Here, we show that NALCN encodes a current that is activated by M3 muscarinic receptors (M3R) in a pancreatic beta-cell line. This current is primar...
